The local DJ and producer, who has received global recognition for his work on Drake's One Dance, has been booked for the prestigious event, where he will share a stage with the likes of Akon, Shaggy and Rich Homie Quan.
The festival, hosted by Akon and dubbed "the first truly international music festival", will be held in Atlanta on Saturday 13 August.
Maphorisa reacted to the news by posting a picture of the line-up, alongside the caption: "Big things #KingOfAfrica"
Fans and friends, including George Avakian, Sbuda Roc and J'Something, all took to the comments section to congratulate Maphorisa on the gig.
Last year, the event raised money for the Akon Lighting Africa Solar Academy in an effort to put electricity in 600 million homes in 14 countries, with organisers set to continue the practice this year.
